Enantioselective hydrogenation of isoquinolines.

چکیده
Asymmetric hydrogenation of (hetero)arenes is one of the most straightforward ways to synthesize enantiomerically pure, saturated or partially saturated cyclic molecules. Recent progress has significantly expanded the substrate scope of this reaction. Whereas a number of highly efficient catalytic systems have been found for the asymmetric hydrogenation of quinolines, structurally related isoquinolines and pyridines are still regarded as challenging substrates for asymmetric hydrogenation (Scheme 1). The difficulty is presumably due

Despite significant progress in the area of asymmetric hydrogenation, the enantioselective hydrogenation of aromatic and heteroaromatic compounds still remains a major challenge. Only a few examples with moderate enantioselectivity, which rely on unique catalyst systems and suffer from a limited scope of suitable substrates, have been described so far. There are several reasons that might expla...

The past decade has witnessed rapid progress in the field of asymmetric hydrogenation of aromatic compounds, a transformation, which is regarded as one of the most straightforward means for accessing enantiopure cyclic compounds. Extensive research has significantly expanded the substrate scope of this reaction, and substrates such as quinolines, quinoxalines, indoles, furans, pyrroles, pyridin...

1,2,3,4-Tetrahydroisoquinolines (THIQs), a class of highly important molecular skeletons abundant in natural alkaloids and biologically active compounds, are often used as key intermediates for the synthesis of pharmaceutical drugs and drug candidates. To date, synthetic efforts have focused on introducing chirality at the C1 position with configurational integrity by employing the following sy...
متن کاملEnantioselective dearomatization of isoquinolines by anion-binding catalysis en route to cyclic α-aminophosphonates.
An enantioselective dearomatization of isoquinolines has been developed using chiral anion-binding catalysis. This transformation, catalyzed by a simple and easy to prepare tert-leucine-based thiourea derivative, makes use of silyl phosphite as a nucleophile and generates cyclic α-aminophosphonates. This is the first time asymmetric anion-binding catalysis has been applied to the synthesis of α...
